Cryo Electron Tomography

I joined the Taylor lab in the middle of 2006. I am working on 3D reconstruction of Insect Flight Muscle (IFM) by electron tomography and following image processing. The high degree order of the IFM makes it an ideal specimen for studying the structure of crossbridges in different states. After acquiring the tilt series on the electron microscopy, the tilt series were repeatedly aligned by cross correlation with a reference calculated from the back projection of the last cycle alignment. And then the crossbridge motifs were extracted from the inverse-Fourier-transformed final map. The motifs were aligned against reference(s) and finally classified by hierarchical ascendant methods.

At the same time, I am working on the cryo-electron tomography of thin margins of cells and isolated cell assemblies.
